从中华鳖Pelodiscussinensis和山瑞鳖Peleasteindachneri的组织材料中提取DNA,扩增约110bp的线粒体12SrRNA基因片段并进行序列分析,结合从Genbank中检索到的缘板鳖Lisemyspunctata的序列,构建了3种鳖的12SrRNA基因片段序列数据库。序列比较的结果表明中华鳖与其它2种鳖的这段序列均有差别。序列差异在11.93%~19.78%之间。从江苏省药品检验所提供的4个鳖甲检品各取样0.1~0.5g提取DNA,扩增与上述相同的基因片段,与构建的数据库进行比较,结果表明4个检品中只有1块为药典规定的正品,其余3块均为混淆品。
DNA was extracted from tissues of Pelodiscussinensis and Peleasteindachneri, and about 110bp mitochondrial 12SrRNA gene fragment was amplified and sequenced. Based on the sequence of Lisemyspunctata retrieved from Genbank, three species of turtles 12SrRNA gene fragment sequence database. Sequence comparison results show that the soft-shelled turtle and other two kinds of turtle in this sequence are different. The sequence differences ranged from 11.93% to 19.78%. 4 samples of turtle shell from the Jiangsu Provincial Institute for Drug Control were sampled from 0.1 to 0.5g to extract the DNA and amplify the same gene fragment as above and compare with the constructed database to find that only 4 of the 4 samples One for the Pharmacopoeia prescribed a genuine, the remaining three are confused products.
