目的:研究p53抑癌基因与大肠癌发生与发展的关系.方法:应用原位分子杂交技术对62例大肠癌区、癌移行区及癌旁分区大肠组织中p53癌基因(p53 mRNA)进行检测.结果:大肠癌区呈 p53 mRNA杂交阳性反应,阳性颗粒主要分布在癌细胞浆内,少部分阳性颗粒核浆共存,阳性癌细胞多呈灶状分布;癌区p53 mRNA杂交阳性率及阳性强度较癌移行区为高(P<0.01),癌旁区为阴性;大肠癌伴有淋巴结及肝转移组,p53 mRNA杂交阳性率明显高于无淋巴结及肝转移组.结论:p53 mRNA表达与大肠癌发生及发展有密切关系,癌组织中 p53 mRNA杂交阳性结果可作为评估大肠癌转移和生存预后的分子生物学指标,对手术切除缘范围评估具有一定临床指导意义.
Objective: To study the relationship between p53 tumor suppressor gene and the occurrence and development of colorectal cancer. Methods: To detect the expression of p53 oncogene (p53 mRNA) in 62 colorectal cancer regions, transitional cancerous tissues, and paraneoplastic tissues by in situ hybridization Results: The colorectal cancer area showed a positive reaction of p53 mRNA hybridization. The positive particles were mainly distributed in the cytoplasm of the cancer cells. A few positive particles had plasmocytoplasmic coexistence. The positive cancer cells mostly had spot distribution; the positive rate and positive rate of p53 mRNA hybridization in the cancer region. It was higher in cancer transition area (P<0.01) and negative in paraneoplastic area. In colorectal cancer group with lymph node and liver metastasis, the positive rate of p53 mRNA hybridization was significantly higher than that in no lymph node and liver metastasis group. Conclusion: p53 mRNA expression and large intestine The occurrence and development of cancer are closely related. The positive result of p53 mRNA hybridization in cancer tissue can be used as a molecular biological indicator to evaluate the metastasis and survival prognosis of colorectal cancer, and it has certain clinical significance for the evaluation of surgical margin size.
