目的:通过建立小鼠帕金森病模型,观察丙戊酸钠(valproate,VPA)对中脑黑质多巴胺神经元及胶质源性神经营养因子(glial-derived neurotrophic factor,GDNF)表达的影响,以期探索丙戊酸钠的神经保护作用机制。方法:采用免疫组织化学方法检测小鼠中脑黑质多巴胺神经元;原位杂交方法检测纹状体、海马、顶叶皮质GDNF的表达。结果:GDNF在模型组和VPA组表达明显增加。结论:VPA可能通过上调GDNF的表达而起到神经保护作用。
OBJECTIVE: To investigate the effect of valproate (VPA) on the expression of dopamine neurons and glial derived neurotrophic factor (GDNF) in the substantia nigra by establishing a mouse Parkinson’s disease model, In order to explore the neuroprotective mechanism of sodium valproate. Methods: The substantia nigra dopamine neurons were detected by immunohistochemical method. The expression of GDNF in striatum, hippocampus and parietal cortex was detected by in situ hybridization. Results: The expression of GDNF in model group and VPA group increased significantly. Conclusion: VPA may play a neuroprotective role by up-regulating the expression of GDNF.
