探讨以调理脾胃气机法组成的中药复方解郁1号对抑郁模型大鼠海马神经细胞元凋亡的影响。方法:将60只SD大鼠随机分为正常组、模型组、西药组及中药组,每组15只,应用孤养加慢性轻度不可预见性应激方法复制大鼠抑郁症模型,采用Open-field法测定各组大鼠的行为变化;TUNEL法检测海马CA3区神经元细胞凋亡情况。结果:模型组大鼠水平活动及垂直活动得分显著减少(P<0.01),海马CA3区内凋亡细胞数显著增加(P<0.01)。与模型组比较,中、西药组大鼠旷场活动次数增加(P<0.01),海马CA3区内凋亡细胞数减少(P<0.01)。结论:解郁1号可改善大鼠抑郁行为,其作用机制可能与抑制海马神经元细胞凋亡有关。
To investigate the effect of Chinese herbal compound Jieyu 1 composed of conditioned spleen-gas machine method on apoptosis of hippocampal neurons in depressive model rats. METHODS: Sixty SD rats were randomly divided into normal group, model group, western medicine group, and Chinese medicine group, 15 rats in each group. The model of depression in rats was duplicated with orphan and chronic mild unpredictable stress. The behavior of rats in each group was measured by the -field method. The neuronal apoptosis in hippocampal CA3 region was detected by TUNEL. Results: The level of activity and vertical activity in the model group decreased significantly (P<0.01), and the number of apoptotic cells in the hippocampal CA3 area increased significantly (P<0.01). Compared with the model group, the number of apoptotic cells in the hippocampal CA3 decreased in the rats in the Chinese and Western medicine groups (P<0.01). Conclusion: Jieyu 1 can improve the depression behavior of rats, and its mechanism may be related to the inhibition of hippocampal neuronal cell apoptosis.
