目的:研究青藤碱对宫颈癌HeLa细胞增殖的影响及其机制。方法:不同浓度青藤碱分别处理体外培养的人宫颈癌细胞系HeLa细胞,采用MTT法检测处理72h后HeLa细胞的增殖,流式细胞仪测定细胞凋亡。Westernblot检测COX-2和Bcl-2蛋白表达情况。结果:0.2、0.4mmol/L青藤碱处理HeLa细胞72h后,细胞增殖明显受到抑制;Westernblot分析表明,中高剂量组COX-2和Bcl-2蛋白表达水平下降;流式细胞仪细胞凋亡分析表明,各剂量组细胞凋亡率较对照组升高。结论:青藤碱在体外能抑制宫颈癌细胞生长,其机制可能与其抑制COX-2和Bcl-2蛋白表达,促进细胞凋亡相关。
Objective: To investigate the effect of sinomenine on the proliferation of cervical cancer HeLa cells and its mechanism. Methods: Human cervical cancer cell line HeLa cells were treated with different concentrations of sinomenine. The proliferation of HeLa cells was detected by MTT assay and the apoptosis was detected by flow cytometry. Western blot analysis of COX-2 and Bcl-2 protein expression. Results: After treated with 0.2, 0.4 mmol / L sinomenine for 72 h, the proliferation of HeLa cells was significantly inhibited. Western blot analysis showed that the expression levels of COX-2 and Bcl-2 were decreased in medium and high dose groups, and apoptosis was detected by flow cytometry The results showed that the apoptotic rate in each dose group was higher than that in control group. Conclusion: Sinomenine can inhibit the growth of cervical cancer cells in vitro, which may be related to the inhibition of COX-2 and Bcl-2 protein expression and the promotion of apoptosis.
