以总黄酮回收率为指标,通过静态吸附和解吸附筛选最佳树脂,确立苦菜地上部分总黄酮的富集纯化工艺。通过静态吸附与解吸附对9种不同极性的大孔树脂进行筛选,优化最佳树脂的上样pH值、上样浓度、上样流速、洗脱流速、洗脱浓度、洗脱体积和径高比等工艺参数。AB-8树脂是富集纯化苦菜地上部分总黄酮的最佳树脂,最佳工艺参数为:上样pH值为pH=5.0,上样液总黄酮质量浓度为0.672 3 mg/mL,吸附流速为2 mL/min,解吸附流速为3 mL/min,乙醇洗脱液的浓度为70%,洗脱体积为2 BV,树脂柱径高比为1∶10,大孔树脂最多重复使用3次,总黄酮纯度倍数为2.43。AB-8型大孔树脂能有效地富集纯化苦菜地上部分总黄酮。
