目的:建立黄蜀葵花总黄酮固体分散体中金丝桃苷的溶出度测定方法。方法:采用HPLC法,建立金丝桃苷的含量测定方法;以金丝桃苷的溶出度为指标,采用单因素试验法,考察溶出方式、溶出介质及转速对金丝桃苷溶出的影响。结果:金丝桃苷在1.140 6~72.998μg·mL~(-1)范围内,与峰面积的积分值呈现良好的线性关系(r=1.000 0),平均加样回收率98.98%,RSD1.82%(n=9);黄蜀葵花总黄酮固体分散体溶出条件为采用浆法,以0.1 mol·L~(-1)盐酸溶液为溶出介质,转速为75 r·min~(-1),取样时间为40 min。结论:所建方法简便、准确、结果可靠,可用于黄蜀葵花总黄酮固体分散体溶出度测定。
Objective: To establish a method for the determination of the dissolution of hyperoside in the solid dispersion of total flavonoids of Abelmoschus. Methods: HPLC method was used to determine the content of hyperoside. The dissolution of hyperoside was used as an index to study the influence of dissolution method, dissolution medium and speed on the dissolution of hyperoside by single factor test. Results: Hyperin in the range of 1.140 6 ~ 72.998μg · mL ~ (-1) showed a good linear relationship with the integral of peak area (r = 1.000 0). The average recovery was 98.98%, RSD1. 82% (n = 9). The dissolution conditions of the total flavonoids of Abelmoschus japonicus were as follows: the pulp was dissolved in 0.1 mol·L -1 hydrochloric acid solution at the speed of 75 r · min -1, Sampling time is 40 min. Conclusion: The method is simple, accurate and reliable. It can be used to determine the dissolution of total flavonoids in Abelmoschus.
