目的:探索中药复方“健脾活血方”中抗内毒素攻击作用的主效应中药及其主要药理作用环节点。方法:采用均匀设计法及尾静脉注射内毒素诱导的大鼠肝损伤模型,以血浆TNF-α含量为指标筛选主效应中药,并再次以该模型验证其药效;进一步检测肝组织CD68、CD14、TLR4、P-IκB蛋白表达及LBP、TLR2、TNF-α、IL-1β、IL-6的mRNA表达。结果:健脾活血方中抗内毒素攻击的主效应中药为枳壳、白芍、泽泻、五味子的组合;主效应中药可显著降低大鼠血浆TNF-α含量;主效应中药可显著降低大鼠肝组织CD68、CD14、TLR4、P-IκB蛋白表达,且其降低肝组织CD68蛋白有优于健脾活血方的优势;主效应中药可显著降低LBP、TLR2、TNF-α、IL-1β、IL-6mRNA表达,均有优于全方的优势。结论:枳壳、白芍、泽泻、五味子组合是健脾活血方中对抗内毒素诱发大鼠急性肝损伤的的主效应中药;主效应中药抗内毒素攻击的主要药理环节在于抑制肝脏库普弗细胞的活化。
OBJECTIVE: To explore the main effect of traditional Chinese medicine and its main pharmacological action points in traditional Chinese medicine prescription “Jianpi Huoxue Recipe” against endotoxin attack. Methods: The model of hepatic injury induced by endotoxin in rat tail vein was established by uniform design method. The main effect of TCM was screened by the level of plasma TNF-α, and the model was used again to verify its efficacy. The levels of CD68, CD14 , TLR4, P-IκB protein expression and LBP, TLR2, TNF-α, IL-1β, IL-6 mRNA expression. Results: The main effect of Jianpi Huoxue Fang against endotoxin attack was the combination of Fructus Aurantii, Radix Paeoniae Alba, Alisma orientalis and Schisandra. The main effect of traditional Chinese medicine could significantly reduce the content of TNF-α in plasma of rats. The expression of CD68, CD14, TLR4, P-IκB protein in rat liver tissue was significantly lower than that of Jianpi Huoxue Decoction. The main effect of traditional Chinese medicine was that LBP, TLR2, TNF-α, IL- IL-6 mRNA expression, all superior to all the advantages. Conclusion: Fructus Aurantii, Radix Paeoniae Alba, Alisma orientalis and Schisandra are the main effects of Jianpi Huoxue Fang against endotoxin-induced acute liver injury in rats. The main pharmacological effects of traditional Chinese medicine against endotoxin attack are inhibition of liver Kupu Activation of euphonism.
