通过考察对血清组胺,Ig E,IL-4,IFN及TNF-α的影响,评价鼻鼽颗粒醇沉工艺的合理性。以咖啡酸和迷迭香酸的含量为综合指标,采用Plackett-Burman设计筛选影响鼻鼽颗粒水提组醇沉工艺的显著性影响因素;在此基础上,通过Box-Behenken设计对显著性因素进一步优化,确定最佳醇沉工艺。确定鼻鼽颗粒水提组的最佳醇沉工艺条件为:浓缩液相对密度1.15(65℃)、醇浓度70%、静止时间12 h。药效学评价结合Plackett-Burman和Box-Behnken设计实验优化确定的醇沉工艺稳定可行,预测性好,为鼻鼽颗粒的产业化生产提供了可靠依据。
By investigating the effects of histamine, Ig E, IL-4, IFN and TNF-α on the serum, the rationality of the nasogastric alcohol precipitation process was evaluated. Taking the contents of caffeic acid and rosmarinic acid as the comprehensive indexes, the Plackett-Burman design was used to screen the significant influencing factors that affect the alcohol precipitation process of the Rhizoma Anemarrhenal particle water extraction group. On this basis, the Box-Behenken design was used to evaluate the significant factors Further optimization, to determine the best alcohol precipitation process. The optimal alcohol precipitation conditions for the water extract of Rhizoma Anemarrhenae were as follows: the relative density of the concentrate was 1.15 (65 ℃), the alcohol concentration was 70% and the rest time was 12 h. Pharmacodynamics evaluation combined with Plackett-Burman and Box-Behnken design experiments to optimize the determination of alcohol precipitation process is stable and predictable, and provides a reliable basis for the industrial production of epistaxis particles.
