目的探讨β-catenin和基质金属蛋白酶-7在HBV相关性肝癌组织中的表达及其相互关系和临床意义。方法经病理证实的HBV阳性HCC组织标本36例、肝血管瘤组织12例、正常肝脏组织12例,免疫组化SP法检测β-catenin和MMP-7的表达情况。结果肝癌组织69.4%(25/36)中可检测到β-catenin异常表达,其中47.2%(17/36)可见胞核染色;肝癌组织β-catenin异常表达率较肝血管瘤和正常肝组织明显升高(P<0.01);在低分化有转移肝癌组织与高分化无转移肝癌组织相比,β-catenin的异常表达率有显著变化(P<0.01)。MMP-7蛋白阳性表达率较肝血管瘤组织及正常肝脏组织明显升高;伴远处转移肝组织MMP-7阳性表达率较无转移肝组织显著升高。肝癌组织中β-catenin的异常表达与MMP-7呈正相关(P<0.01)。结论β-catenin在HBV相关性肝癌组织中的异常表达可上调MMP-7的表达,与肝癌的发生、发展及转移有关。
Objective To investigate the expression of β-catenin and matrix metalloproteinase-7 in HBV-related hepatocellular carcinoma and its relationship and clinical significance. Methods Thirty-six HCC tissues were confirmed by pathology, including 12 cases of hemangiomas and 12 cases of normal liver tissues. The expressions of β-catenin and MMP-7 were detected by immunohistochemical SP method. Results Abnormal expression of β-catenin was detected in 69.4% (25/36) of the HCC tissues, of which 47.2% (17/36) showed nuclear staining. The abnormal expression of β-catenin in HCC tissues was significantly higher than that in hepatic hemangiomas and normal liver tissues (P <0.01). The abnormal expression rate of β-catenin in poorly differentiated metastatic HCC tissues was significantly higher than that in well-differentiated and non-metastatic HCC tissues (P <0.01). The positive expression rate of MMP-7 protein was significantly higher than that of hepatic hemangiomas and normal liver tissues. The positive expression rate of MMP-7 in distant metastasis liver tissues was significantly higher than that in non-metastatic liver tissues. The abnormal expression of β-catenin in hepatocellular carcinoma was positively correlated with MMP-7 (P <0.01). Conclusion Aberrant expression of β-catenin in HBV-related HCC tissue may up-regulate the expression of MMP-7, which may be related to the occurrence, development and metastasis of HCC.
