目的探讨自噬调节对胶质瘤起始细胞辐射敏感性的影响。方法收集手术切除的胶质瘤标本,制备胶质瘤起始细胞系SU-2,用雷帕霉素处理后再行X线照射(A组)或直接行X线照射(B组)。MTT比色法和克隆形成实验观察SU-2细胞增殖情况;应用Hoechst 33258荧光染色检测SU-2细胞凋亡。结果 A组的细胞增殖率和克隆形成率较B组明显降低(P<0.01),细胞凋亡率显著增加。结论胶质瘤起始细胞经自噬激动剂雷帕霉素处理后,对X线照射的辐射敏感性明显增加。
Objective To investigate the effect of autophagy on the radiosensitivity of glioma-initiating cells. Methods Surgical resected glioma specimens were collected and the glioma cell line SU-2 was prepared. After treatment with rapamycin, X-ray irradiation (group A) or direct X-ray irradiation (group B) was performed. The proliferation of SU-2 cells was observed by MTT assay and colony formation assay. The apoptosis of SU-2 cells was detected by Hoechst 33258 staining. Results The cell proliferation rate and colony formation rate of group A were significantly lower than those of group B (P <0.01), and the apoptosis rate of group A was significantly increased. Conclusion The radiosensitivity of glioma initiating cells to X-ray irradiation after autophagy agonist rapamycin is significantly increased.
