目的:探讨大鼠肾移植急性排异反应中Fas基因的表达与细胞凋亡的关系。方法:建立大鼠同种异体肾移植模型,应用免疫组织化学方法检测Fas基因的表达;应用原位DNA片段未端标记法检测移植肾组织细胞凋亡状态.结果:正常大鼠肾Fas基因主要呈弱阳性或阴性表达,而移植肾Fas基因表达显著增强,且与肾曲小管细胞凋亡指数呈显著的正相关关系(rs=0.512,P<0.001).结论:大鼠移植肾Fas基因的过量表达可引起肾组织细胞的凋亡,导致移植肾结构的破坏和功能的丧失.
Objective: To investigate the relationship between Fas gene expression and apoptosis in acute rejection of renal allograft in rats. Methods: Rat renal allograft model was established. The expression of Fas gene was detected by immunohistochemical method. Apoptosis status of renal allograft tissue was detected by in situ DNA fragment end labeling. Results: The expression of Fas gene in kidney of normal rats was mainly weakly positive or negative, while the expression of Fas gene in renal allografts was significantly increased. There was a significant positive correlation between the expression of Fas gene and renal tubular cells (rs = 0.512, P <0 .001). Conclusion: The overexpression of Fas gene in rat renal allografts can induce the apoptosis of renal cells, resulting in the destruction of the renal structure and the loss of function.
