已知体外依赖小鼠脾细胞产生抗羊红细胞抗体过程中,组氨酸H_2受体阻断剂可使抗原特异的抗体产生细胞增加。本文在人末梢血淋巴细胞产生抗羊红细胞抗体过程中,添加特异性较高的H_2受体阻断剂famoti-dine,观察其对抗原特异的抗体产生的影响。将人末梢血单核细胞(10×10~6个)培养8天,并同时添加作为抗原的羊红细胞(5×10~6个),另将不加羊红细胞组为对照组。在培养液中加入浓度分别为0.02、0.2、2、20和200μg/ml 的famotidine,培养8天后将培养细胞洗净,与羊红细胞、作为补
It is known that histidine H 2 receptor blockers increase the number of antigen-specific antibody-producing cells in vitro dependent on the production of anti-sheep erythrocyte antibodies by mouse spleen cells. In this study, we used famoti-dine, a specific H 2 receptor blocker, to produce anti-sheep erythrocytic antibodies during the production of anti-sheep erythrocytes by peripheral blood lymphocytes. Human peripheral blood mononuclear cells (10 × 10 ~ 6) were cultured for 8 days. Meanwhile, sheep red blood cells (5 × 10 ~ 6) were added as antigen, and the other group without sheep’s blood cells as control group. Famotidine was added to the culture broth at concentrations of 0.02, 0.2, 2, 20 and 200 μg / ml, respectively. After culturing for 8 days, the cultured cells were washed and incubated with sheep erythrocytes
