通过优化预处理和检测方法,采用Waters C18反相色谱柱,以磷酸缓冲盐和乙腈为流动相,外标法测定GA3的回收率为78.42%,变异系数±3.9,保留时间稳定,首次建立了毛竹种子中植物内源激素GA3含量的测定方法。运用此法测定毛竹种子萌发过程中GA3含量的变化,结果表明,从浸种到出苗时GA3的含量在2.56~14.75μg.g-1,吸胀阶段含量变化不明显,而浸种36 h后含量大幅升高,在72 h和即将萌出胚根时达到峰值,随着毛竹种子的萌发进程,GA3含量呈总体上升趋势。
The optimum conditions of pretreatment and detection were established on a Waters C18 reversed-phase column with phosphate buffered saline and acetonitrile as the mobile phase. The recovery of GA3 was 78.42% by external standard method and the coefficient of variation was ± 3.9. Determination of Endogenous Hormones GA3 in Moso Bamboo Seeds. The results showed that GA3 content was 2.56-14.75μg.g-1 from soaking to germination, while the content of GA3 was not significantly changed during the imbibition stage. However, the content of GA3 was significantly increased after soaking for 36 h Which reached its peak at 72 h and immediately before emergence. The GA3 content showed an overall upward trend with the germination process of Phyllostachys pubescens seeds.
