目的:观察苦参碱(matrine)对体外培养的人肉瘤细胞系MS0812细胞的增殖和诱导调亡作用。方法:苦参碱处理人肉瘤细胞系MS0812后,应用MTT比色法检测不同浓度苦参碱和不同作用时间对MS0812细胞的增殖作用的影响,TUNEL法和流式细胞仪检测细胞凋亡。结果:苦参碱浓度大于500.0 mg·L-1作用24h对MS0812细胞增殖有抑制作用;通过TUNEL试剂盒可以发现凋亡阳性细胞;流式细胞仪检测随着苦参碱浓度的增加,凋亡率逐渐升高。结论:苦生碱体外对肿瘤细胞系MS0812的生长抑制和诱导凋亡作用有限。
OBJECTIVE: To observe the effects of matrine on the proliferation and apoptosis of human sarcoma cell line MS0812 cultured in vitro. Methods: After matrine treatment of human sarcoma cell line MS0812, MTT colorimetric assay was used to detect the effect of different concentrations of matrine and different time on the proliferation of MS0812 cells. TUNEL and flow cytometry were used to detect apoptosis. Results: The effect of matrine over 500.0 mg · L-1 for 24 h could inhibit the proliferation of MS0812 cells. Apoptosis-positive cells could be detected by TUNEL kit. Flow cytometry was used to detect apoptosis with increasing concentration of matrine The rate gradually increased. Conclusion: Alkaloids can inhibit the growth and induce apoptosis of MS0812 cell line in vitro.
