目的:探讨羧基肽酶E(CPE)在子宫颈癌组织中表达及临床意义。方法:选取2011年1月至2012年1月在郑州大学第五附属医院择期行手术切除的子宫颈癌患者76例,同期,选取子宫颈上皮内瘤变(CIN)患者53例,以及子宫良性病变患者正常子宫颈组织40例作为对照组,利于实时荧光定量聚合酶链式反应(PCR)技术检测各组子宫颈病变组织中CPE基因表达,利用Kaplan-Meier生存分析法分析子宫颈癌组织中CPE信使核糖核酸(m RNA)相对表达量对患者生存时间的影响。结果:子宫颈癌患者组织中CPE m RNA相对表达量(1.92±0.23),均高于CIN患者的(1.66±0.20)和对照组的(1.18±0.14),差异具有统计学意义(P<0.05);CIN患者中,CINⅡ~Ⅲ患者组织中CPE m RNA相对表达量为(1.76±0.19),高于CINⅠ患者的(1.45±0.13),差异具有统计学意义(P<0.05),即子宫颈癌患者>CINⅡ~Ⅲ患者>CINⅠ患者>对照组;子宫颈癌患者组织中CPE m RNA相对表达量与组织学分级、FIGO 2000分期、淋巴结转移相关(P<0.05);以CPE m RNA相对表达量的P25值为界值,将患者分为低表达组和高表达组,Kaplan–Meier生存分析显示,低表达组患者平均生存时间45.35个月,高表达组患者平均生存时间32.08个月,Log Rank检验显示,差异具有统计学意义(P<0.05)。结论:CPE基因子宫颈癌组织中呈高表达,有望成为子宫颈癌患者早期诊断及预后判断的潜在标志物。
Objective: To investigate the expression of carboxypeptidase E (CPE) in cervical cancer and its clinical significance. Methods: A total of 76 patients with cervical cancer undergoing elective resection at the Fifth Affiliated Hospital of Zhengzhou University from January 2011 to January 2012 were enrolled in this study. Fifty-three patients with cervical intraepithelial neoplasia (CIN) 40 cases of normal cervical lesions in patients with lesions as a control group, which is conducive to real-time fluorescent quantitative polymerase chain reaction (PCR) detection of cervical lesions in each group of CPE gene expression, Kaplan-Meier survival analysis of cervical cancer Effect of relative expression of CPE messenger RNA (RNA) on survival time of patients. Results: The relative expression of CPE m RNA in cervical cancer patients was 1.92 ± 0.23, which was significantly higher than that in CIN patients (1.66 ± 0.20) and 1.18 ± 0.14 (P <0.05), respectively ). In CIN patients, the relative expression of CPE m RNA in CINⅡ-Ⅲ patients was (1.76 ± 0.19), which was higher than that in CINⅠ patients (1.45 ± 0.13), the difference was statistically significant (P <0.05) Cancer patients> CINⅡ-Ⅲpatients> CINⅠpatients> control group. The relative expression of CPE m RNA in cervical cancer patients was correlated with histological grade, FIGO 2000 stage and lymph node metastasis (P <0.05) Kaplan-Meier survival analysis showed that the average survival time of patients with low expression group was 45.35 months, and the average survival time of high expression group patients was 32.08 months. Log Rank test showed that the difference was statistically significant (P <0.05). Conclusion: The high expression of CPE gene in cervical cancer tissue is expected to become a potential marker for early diagnosis and prognosis of cervical cancer patients.
