本文分离纯化了正常大白鼠肝脏和癌前病变大白鼠肝脏组织线粒体谷胱甘肽S-转移酶(GST),对其部分性质进行了探讨。经O-(二乙胺基乙基)-纤维素、羟基磷灰石,羧甲基-琼脂糖6B(CM-Sapharose 6B)柱层析以及等电聚焦电泳法,从正常肝组织和癌前病变肝组织中,分离纯化了两种不同性质的GST,暂命名为线粒体GST_1和线粒体GST_2。通过十二烷基磺酸钠(SDS)-聚丙烯酰胺凝胶电泳法和等电聚焦电泳法测定的线粒体GST_1的分子量约为23000,等电点为6.2,线粒体CST_2的分子量约为45000,等电点为9.1。在正常组织和癌前病变组织中CST同工酶无质上的差异。但是,线粒体GST_1在癌前病变组织中的活性明显著高于正常对照组(P<0.01)。
In this paper, we isolated and purified mitochondrial glutathione S-transferase (GST) from the liver of normal rats and precancerous lesioned rat liver, and discussed some of its properties. Chromatography was performed on normal liver tissue and precancerous lesions using O- (diethylaminoethyl) -cellulose, hydroxyapatite, CM-Sepharose 6B, and isoelectric focusing electrophoresis In diseased liver tissue, two different types of GST were isolated and purified, temporarily named mitochondrial GST_1 and mitochondrial GST_2. The molecular weight of mitochondrial GST_1 determined by sodium dodecyl sulfate (SDS) -polyacrylamide gel electrophoresis and isoelectric focusing electrophoresis was about 23000, the isoelectric point was 6.2, the molecular weight of mitochondrial CST_2 was about 45000, etc. The electric point is 9.1. There are no qualitative differences in CST isozymes between normal and precancerous lesions. However, the activity of mitochondrial GST_1 in precancerous lesions was significantly higher than that in the normal control group (P <0.01).
