培养后呈汇合成片的人脐静脉内皮细胞在体外经~(60)Coγ线照射,剂量从0~50Gy。辐照后于不同时间测定内皮细胞内、细胞膜表面和细胞培养上清液中血栓调节蛋白含量。辐照后24h,内皮细胞释放的血栓调节蛋白和von Willebrand因子增高,同时内皮细胞膜表面血栓调节蛋白的分子数和活性也都增高,其增加幅度与照射剂量有关。辐照后2~6d,内皮细胞血栓调节蛋白的合成和释放都降低。揭示了核辐照可引起内皮细胞损伤,血栓调节蛋白可作为核辐照引起内皮细胞损伤的一个标志。对核辐照后内皮细胞功能异常与急性放射病的发病机理之间的关系亦作了讨论。
After cultured, the human umbilical vein endothelial cells were collected and irradiated with ~ (60) Coγ ray in vitro at the dose of 0 ~ 50Gy. After irradiation, the content of thrombomodulin in the endothelial cells, the cell membrane surface and the cell culture supernatant was determined at different times. 24h after irradiation, thrombomodulin and von Willebrand factor released by endothelial cells increased, meanwhile, the molecular number and activity of thrombomodulin on the surface of endothelial cells also increased. The increase range was related to the irradiation dose. 2 ~ 6 days after irradiation, the synthesis and release of endothelial cell thrombomodulin decreased. Revealed that nuclear irradiation can cause endothelial cell damage, thrombomodulin as a marker of endothelial cell damage caused by nuclear irradiation. The relationship between endothelial dysfunction after nuclear irradiation and the pathogenesis of acute radiation sickness was also discussed.
