目的探讨植物生长调节剂赤霉素对大鼠肝细胞线粒体呼吸功能的影响。方法 24只SD雄性大鼠随机分为4组,即对照组和3个赤霉素染毒组,各组动物分别用含0、40、160和640mg/kg赤霉素饲料喂养35天。染毒结束24h后处死动物,取大鼠肝,提取线粒体,分别用荧光分光光度法、紫外分光光度法、Clark氧电极测定线粒体膜电位变化、线粒体呼吸链复合体活性和线粒体呼吸功能的变化。结果高浓度赤霉素处理组大鼠肝细胞线粒体ComplexⅠ和ComplexⅢ活性以及中、高浓度赤霉素处理组肝细胞线粒体ComplexⅣ活性显著高于对照组(P<0.05)。高浓度赤霉素处理组大鼠肝细胞线粒体Ⅲ态耗氧量与对照组相比较差异有统计学意义(P<0.05)。结论动物摄入高浓度赤霉素饲料可使肝细胞线粒体呼吸功能增强。
Objective To investigate the effect of gibberellin on mitochondrial respiratory function in rat hepatocytes. Methods Twenty-four SD male rats were randomly divided into 4 groups: control group and 3 gibberellins groups. The animals in each group were fed with gibberellic acid (0, 40, 160 and 640 mg / kg) for 35 days respectively. Animals were sacrificed 24 hours after the end of the treatment. Rat liver was harvested and mitochondria were extracted. Changes in mitochondrial membrane potential, mitochondrial respiratory chain activity and mitochondrial respiratory function were measured by fluorescence spectrophotometry, UV spectrophotometry and Clark oxygen electrode. Results The activity of mitochondria ComplexⅠ and ComplexⅢ in hepatocytes of high concentration gibberellic acid treatment group and the activity of mitochondrial ComplexⅣ of hepatocytes in medium and high concentration gibberellic acid treatment group were significantly higher than those in control group (P <0.05). Compared with the control group, the oxygen consumption of mitochondrial III of hepatocytes in high concentration gibberellic acid treatment group was significantly different (P <0.05). Conclusion High intake of GA3 in animals can enhance mitochondrial respiratory function of hepatocytes.
