目的:探讨影响老年糖尿病周围血管病变患者(PAD)行外周血干细胞动员采集效果的因素。方法:对91例老年PAD患者行粒细胞集落刺激因子(G-CSF)动员后,采用Cobe Spectra血细胞分离机行自体外周血干细胞(PBSC)采集。PBSC采集后取混匀后的采集物少量悬液计数单个核细胞数(MNC)、CD34+细胞数。采用多元线性回归分析,G-CSF剂量、采集前白细胞计数、体重指数(BMI)、糖化血红蛋白(HbA1c)、甘油三酯(TG)和低密度脂蛋白(LDL-C)等因素对采集物MNC和CD34+细胞数的影响。结果:BMI、TG对采集的MNC数有影响;HbA1c、LDL-C与CD34+呈负相关;动员剂的剂量、采集前白细胞计数与MNC总量和CD34+细胞数量关系不密切。结论:根据不同病情制定个体化动员方案,减少不利因素,老年PAD患者也可安全、成功地行PBSC采集。
Objective: To investigate the factors influencing the mobilization and collection of peripheral blood stem cells in elderly patients with diabetic peripheral vascular disease (PAD). Methods: Ninety elderly PAD patients were mobilized with granulocyte colony-stimulating factor (G-CSF) and collected by using peripheral blood stem cells (PBSC) using a Cobe Spectra blood cell separator. Take a small amount of collected suspension collected after PBSC collection count the number of mononuclear cells (MNC), CD34 + cells. Multivariate linear regression analysis, G-CSF dose, white blood cell count, body mass index (BMI), HbA1c, triglyceride (TG) and low density lipoprotein (LDL- And the number of CD34 + cells. Results: BMI and TG had an influence on the number of MNC collected; HbA1c and LDL-C were negatively correlated with CD34 +; the dose of mobilization agent and pre-collection leukocyte count were not closely related to the total amount of MNC and CD34 + cells. Conclusion: According to different conditions to develop individualized mobilization program to reduce adverse factors, elderly patients with PAD can safely and successfully collected PBSC line.
