目的建立罗勒中芦丁和槲皮素的含量测定方法,并对不同产地药材进行含量测定。方法采用高效液相色谱法将罗勒中芦丁和槲皮素进行测定;芦丁的流动相为乙腈-0.4%磷酸(18∶82),槲皮素的流动相为乙腈-0.4%磷酸(32∶68);检测波长均为360 nm;流速均为1.0 mL·min-1;柱温均为30℃。结果芦丁的线性范围为4.95~24.75μg(r=0.999 7),平均回收率为100.7%,RSD=1.62%(n=6)。槲皮素的线性范围为1.792~8.064μg(r=0.999 7),平均回收率为98.3%,RSD为0.94%(n=6)。不同产地罗勒药材中芦丁和槲皮素的含量分别在0.39~0.71 mg·g-1和0.29~0.58 mg·g-1之间。结论本法专属性强,重复性好,结果准确可靠,可作为罗勒的质量控制方法。
Objective To establish a method for the determination of rutin and quercetin in basil, and to determine the contents of rutin and quercetin in different producing areas. Methods Rutin and quercetin in basil were determined by high performance liquid chromatography. The mobile phase of rutin was acetonitrile-0.4% phosphoric acid (18:82), the mobile phase of quercetin was acetonitrile-0.4% phosphoric acid : 68). The detection wavelength was 360 nm and the flow rate was 1.0 mL · min-1. The column temperature was 30 ℃. Results The linear range of rutin was 4.95 ~ 24.75μg (r = 0.999 7), with an average recovery of 100.7% and RSD = 1.62% (n = 6). The linear range of quercetin was 1.792 ~ 8.064μg (r = 0.999 7) with the average recovery of 98.3% and RSD 0.94% (n = 6). The contents of rutin and quercetin in the basil of different origins were between 0.39 ~ 0.71 mg · g-1 and 0.29 ~ 0.58 mg · g-1, respectively. Conclusion This method is special, reproducible and accurate. It can be used as a quality control method for basil.
