目的:对黄独试管苗微型块茎的诱导形成进行初步研究,为黄独微型块茎的工厂化生产及种质资源保存提供理论依据。方法:采用植物组织培养技术和单因子试验的方法,以黄独试管苗的带芽茎段为外植体,研究糖类、无机盐、培养方式、活性炭以及带芽茎段生理状态等因子对黄独微型块茎诱导形成的影响。结果:黄独微型块茎诱导的最佳糖类为60 g/L蔗糖或90 g/L白砂糖;黄独微型块茎诱导的最佳无机盐浓度为MS;黄独微型块茎诱导的最佳培养方式为固-液双层培养;活性炭对黄独微型块茎诱导有显著的促进作用,其最佳浓度为0.03%;带芽茎段成熟度越高,形成微型块茎的时间越短。结论:初步建立了黄独微型块茎的快速诱导方法,为黄独微型块茎在农业生产上的应用提供了新途径。
OBJECTIVE: To study the formation and induction of mini tuber of yellow detached plantlets in vitro, and to provide a theoretical basis for the industrialization and conservation of yellow miniature tubers. Methods: Plant tissue culture technology and single factor test were used to study the effects of carbohydrate, inorganic salt, culture method, activated carbon and the physiological status of stems and shoots with yellow buds Effect of yellow miniature tuber inducement formation. Results: The best sugars induced by yellow miniature tuber was 60 g / L sucrose or 90 g / L white sugar. The best inorganic salt concentration induced by yellow miniature tuber was MS. Liquid-solid bilayer culture. Activated charcoal had a significant promotion effect on yellow tubercle tuber induction, with the optimal concentration of 0.03%. The higher the maturity of bud stem was, the shorter the time of forming mini tuber was. Conclusion: The rapid induction method of yellow miniature tubers was established preliminarily, which provided a new way for the application of yellow miniature tubers in agricultural production.
