高效毛细管电泳(HighPerformanceCapillaryElectrophoresis,简称HPCE))技术近十年来得到了迅速的发展。由于它具有进样量小(nl级)、灵敏度高(10-10mol)、分辨率为百万理论塔板数(高出HPLC数十倍),而且可以快速、高效地分离分子量达106-1011道尔顿的生物大分子等特点,因此受到了化学、生命科学及药物分析、环保等领域的极大量视[1~8]。目前,国外已出现了近十种商品化仪器,而本文介绍的新型毛细管电泳仪是我国第一台商品化仪器,采取了两种进样方法、两种毛细管冲洗方式、多波长UV检测、恒温及微机数据处理系统。本文简要说明了这种仪器的原理、结构和特征,并给出了分离蛋白质、核苷酸及单糖的实验条件和图谱。
HighPerformanceCapillary Electrophoresis (abbreviated as HPCE) technology has been rapidly developed in the past decade. Due to its small sample volume (nl), high sensitivity (10-10 mol), resolution of millions of theoretical plates (several tens of times higher than HPLC), and the ability to separate molecular weights up to 106-1011 quickly and efficiently. Due to Dalton’s biological macromolecules, it has received a great deal of attention in the fields of chemistry, life sciences, drug analysis, and environmental protection [1-8]. At present, nearly ten commercial instruments have appeared in foreign countries. The new capillary electrophoresis apparatus introduced in this paper is the first commercially available instrument in China. Two kinds of sample injection methods, two types of capillary flushing, multi-wavelength UV detection, and constant temperature have been adopted. And computer data processing system. This article briefly describes the principle, structure and characteristics of this instrument, and gives experimental conditions and maps for the separation of proteins, nucleotides and monosaccharides.
