目的:探讨鞘内阿米洛利合用吗啡对大鼠骨癌的镇痛作用。方法:将walker 256乳腺癌细胞注入SD雌性大鼠胫骨内形成骨癌痛模型,再行蛛网膜下腔置管。随机将大鼠分成对照组、阿米洛利组、吗啡组、混合药物组。鞘内给药前后测定大鼠机械性缩足反应阈值。采用线性回归的方法计算各药物的半数有效剂量(ED_(50))及95%的可信区间(CI_(95))。使用Isobolographic评价药物的相互作用。结果:鞘内单独注射阿米洛利可产生剂量依赖性的抗骨癌痛作用,其半数有效剂量(ED_(50))及95%可信区间CI_(95)分别是:63.8μg和53.2~74.5μg。鞘内阿米洛利和吗啡合用可以产生协同镇痛作用。结论:鞘内单独注射阿米洛利、吗啡可以产生明显剂量依赖性的抗骨癌痛作用;鞘内注射阿米洛利可以增强吗啡的抗骨癌痛作用。
Objective: To investigate the analgesic effect of intrathecal amiloride combined with morphine on rat bone cancer. METHODS: Walker 256 breast cancer cells were injected into the tibia of female SD rats to form a model of bone cancer pain. The subarachnoid space was then cannulated. Rats were randomly divided into control group, amiloride group, morphine group, mixed drug group. Before and after intrathecal administration rat mechanical contraction reaction threshold. The median effective dose (ED_ (50)) and 95% confidence interval (CI_ (95)) of each drug were calculated by linear regression. Isobolographic was used to evaluate drug interactions. RESULTS: Intrathecal injection of amiloride alone resulted in a dose-dependent anti-cancer effect. The median effective dose (ED_ (50)) and 95% confidence interval CI 95 were 63.8 μg and 53.2 ~ 74.5 μg. Intrathecal amiloride and morphine can produce a synergistic analgesic effect. Conclusion: Intrathecal injection of amiloride alone and morphine can produce a significant dose-dependent anti-cancer effect of bone cancer. Intrathecal injection of amiloride can enhance the anti-cancer effect of morphine.
