目的研究重度支原体肺炎患儿感染模式识别受体-黑色素瘤分化相关基因5(MDA5)、视磺酸可诱导基因-1(RIG-1)蛋白表达,旨在分析两者在支原体肺炎感染中的价值,从而早期诊断、评价病情。方法选取2013年5月-2016年12月该院重度支原体肺炎感染患儿50例作为观察组,与同期选取健康体检儿童50例作为对照组。采用酶联免疫吸附法(ELISA)检测两组研究对象MDA5和RIG-1蛋白表达。结果观察组急性期和恢复期MDA5、RIG-1蛋白表达分别达(15.16±3.86)ng/ml、(12.42±3.11)ng/ml和(11.48±2.51)ng/ml、(5.81±2.33)ng/ml,均高于对照组的(6.37±2.06)ng/ml和(2.68±1.35)ng/ml,差异均有统计学意义(t=14.24、20.30;13.11、9.06,均P<0.05)。且观察组急性期MDA5和RIG-1蛋白表达高于观察组恢复期,差异有统计学意义(t=11.15、8.22,均P<0.05)。MDA5、RIG-1表达Spearman相关性分析显示两者蛋白表达呈显著正相关性(r=0.63,P<0.01)。结论模式识别受体的MDA5和RIG-1表达可作为早期诊断重度支原体肺炎感染及评价病情的重要指标,加强两者的检测有助于及时掌握病原菌入侵状况和组织稳态信息,及时采取靶向治疗杀灭病原菌、恢复机体稳态,协作患儿度过危险期。
Objective To study the expression of pattern recognition receptors-melanoma differentiation-related gene 5 (MDA5) and retinoic acid-inducible gene-1 (RIG-1) in children with severe mycoplasma pneumonia and to analyze the relationship between them in mycoplasma pneumoniae infection Value, so early diagnosis, evaluation of the disease. Methods From May 2013 to December 2016, 50 children with severe mycoplasma pneumoniae infection were selected as the observation group and 50 healthy children were selected as the control group. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression of MDA5 and RIG-1 protein in two groups. Results The expression of MDA5 and RIG-1 protein in acute and convalescent groups were (15.16 ± 3.86) ng / ml, (12.42 ± 3.11) ng / ml and (11.48 ± 2.51) ng / ml, / ml were higher than those in the control group (6.37 ± 2.06) ng / ml and (2.68 ± 1.35) ng / ml, respectively, with statistical significance (t = 14.24,20.30; 13.11,9.06, all P <0.05). The expression of MDA5 and RIG-1 protein in the acute phase of the observation group was higher than that of the observation group (t = 11.15, 8.22, all P <0.05). Spearman correlation analysis of MDA5 and RIG-1 expression showed a significant positive correlation (r = 0.63, P <0.01). Conclusions The expression of pattern recognition receptors, MDA5 and RIG-1, can be used as an important indicator for the early diagnosis of severe mycoplasma pneumonia infection and evaluation of the disease.
