目的:探讨NGX6mRNA两种转录本NGX6-S和NGX6-L在结直肠癌中的表达及功能。方法:采用原位杂交法检测NGX6-L和NGX6-S在不同分期结直肠癌癌组织及其配对正常组织中的表达特征,并分析其与血清癌胚抗原(CEA)的相关性。结果:在结直肠癌组织中短转录本NGX6-S的表达明显高于长转录本NGX6-L(P=0.008)。NGX6-S和NGX6-L在不同分期结直肠癌癌组织中的表达差异均无统计学意义(P>0.05)。NGX6-S在Duke A,B和C三期结直肠癌组织中的表达与其在配对正常组织的表达差异无统计学意义(P>0.05),在有远处转移的Duke D期结直肠癌组织中的表达低于配对正常组织(P=0.033)。NGX6-L在Duke A,B,C和D期结直肠癌组织中的表达与配对正常组织差别无统计学意义(P>0.05)。结直肠癌中NGX6-S,NGX6-L的表达与血清CEA浓度无相关性。结论:在结直肠癌中NGX6基因发挥抑瘤功能的主要形式是短转录本NGX6-S,其表达下调可能与结直肠癌的远处转移相关。
Objective: To investigate the expression and function of two NGX6 mRNA transcripts NGX6-S and NGX6-L in colorectal cancer. Methods: The expression characteristics of NGX6-L and NGX6-S in different stages of colorectal cancer tissues and matched normal tissues were detected by in situ hybridization, and their correlation with serum carcinoembryonic antigen (CEA) was analyzed. Results: The expression of short transcript NGX6-S in colorectal cancer tissues was significantly higher than that of the long transcript NGX6-L (P = 0.008). There were no significant differences in the expression of NGX6-S and NGX6-L in different stages of colorectal cancer (P> 0.05). The expression of NGX6-S in Dukes A, B and C stage 3 colorectal cancer tissues was not significantly different from that in paired normal tissues (P> 0.05). In Duke D stage colorectal cancer tissues with distant metastasis The expression was lower in matched normal tissues (P = 0.033). The expression of NGX6-L in Duke A, B, C and D colorectal cancer tissues was not significantly different from the matched normal tissues (P> 0.05). There was no correlation between the expression of NGX6-S and NGX6-L in colorectal cancer and serum CEA concentration. CONCLUSION: The main form of NGX6 gene inhibiting tumor function in colorectal cancer is the short transcript NGX6-S, which may be related to the distant metastasis of colorectal cancer.
