目的探讨MDM2(murine double minute 2,MDM2)表达与胃癌转移的关系。方法采用免疫组织化学染色方法检测94例胃腺癌组织中MDM2的表达水平,分析MDM2表达与胃癌转移间的关系;应用细胞实验进一步探讨下调MDM2表达对胃癌细胞转移和侵袭能力的影响。结果免疫组织化学染色结果显示MDM2高表达与胃癌转移有关,而利用细胞实验证实,下调MDM2表达可显著抑制胃癌细胞的转移和侵袭能力,其与胃癌细胞p53状态无关。结论 MDM2高表达可通过非p53途径促进胃癌细胞转移。
Objective To investigate the relationship between the expression of murine double minute 2 (MDM2) and gastric cancer metastasis. Methods Immunohistochemical staining was used to detect the expression of MDM2 in 94 cases of gastric adenocarcinoma. The relationship between the expression of MDM2 and the metastasis of gastric cancer was analyzed. The effect of down-regulating MDM2 expression on the metastasis and invasion ability of gastric cancer cells was also investigated by cell experiment. Results The results of immunohistochemistry showed that MDM2 overexpression was associated with gastric cancer metastasis. However, the down-regulation of MDM2 expression could significantly inhibit the metastasis and invasion ability of gastric cancer cells, which was not related to the p53 status of gastric cancer cells. Conclusion MDM2 overexpression promotes gastric cancer cell metastasis through non-p53 pathway.
