为探索诱导免疫耐受的途径和方法,作者采用大鼠异位心脏(颈部)移植模型,经外周途径输注不同品系供者的不同特异性抗原,包括供体特异性输血( D S T)、供体特异性脾细胞( D S S L)、供体特异性骨髓细胞( D S B M)。结果显示: D S T、 D S S L和 D S B M 均能诱导受者对移植物产生特异性免疫耐受,其中以脾细胞和骨髓细胞效果最好,平均移植物存活时间分别达到 429±667 和 403±116 天。混合淋巴细胞反应( M L R)和 T 亚群( C D8+ )检测在一定程度上反映了机体 T 细胞免疫状态,有助于对耐受的评价。成分输注供者特异性抗原作为刺激抗原,可望成为较理想的临床耐受诱导途径。
In order to explore ways and methods of inducing immune tolerance, the authors used rat heterotopic heart (neck) transplantation models to transfuse different specific antigens from different lineage donors via peripheral routes, including donor-specific transfusion (DST ), Donor-specific spleen cells (D S S L), donor-specific bone marrow cells (D S B M). The results showed that DST, DsS and DsBM could induce recipients to produce specific immune tolerance to the grafts, and the spleen cells and myeloid cells were the best and the mean graft survival time was 42 9 ± 6 67 and 40 3 ± 11 6 days. MLC and T subpopulations (C D8 +) test, to a certain extent, reflect the T cell immune status and contribute to the evaluation of tolerance. Component infusion of donor-specific antigen as a stimulus antigen, is expected to become more ideal clinical tolerance induction pathway.
