目的:探讨抗凋亡基因SURVIVIN及其异构体和天然互补序列EPR-1在急性白血病(AL)中的表达, 以及在AL的发生、预后等方面的意义。方法:采用RT-PCR方法检测86例初治AL患者SURVIVIN及其异构体SUR- VIVIN-2B、SURVIVIN-EX3、EPR-1基因的MRNA表达。结果:(1)本组AL患者SURVIVIN MRNA表达率为79.1%,高于正常对照组(31.3%)(P<0.01);(2)SURVIVIN阳性患者完全缓解率(CR)明显低于SURVIVIN阴性组(P<0.01);(3)AL 中存在SURVIVIN-2B、SURVIVIN-△EX3异构体分子;(4)EPR-1在正常人及大多数AL中均有表达,AL以SURVIVIN+/ EPR-1+表达模式为主。结论:在AL中SURVIVIN高表达且与预后有关,并以SURVIVIN+/EPR-1+表达模式为多见; SURVIVIN-2B、SURVIVIN-△EX3可能参与SURVIVIN介导的凋亡调控。
Objective: To investigate the expression of anti-apoptotic gene SURVIVIN, its isoform and natural complementary sequence EPR-1 in acute leukemia (AL) and its significance in the pathogenesis and prognosis of AL. Methods: MRNA expression of SURVIVIN and its isoforms SUR-VIVIN-2B, SURVIVIN-EX3 and EPR-1 gene were detected by RT-PCR in 86 newly diagnosed AL patients. Results: (1) The expression rate of SURVIVIN MRNA in AL patients was 79.1%, higher than that in normal controls (31.3%) (P <0.01); (2) The complete remission rate (CR) (3) SURVIVIN-2B and SURVIVIN- △ EX3 isomers exist in AL; (4) EPR-1 is expressed in both normal and most ALs , AL mainly SURVIVIN + / EPR-1 + expression pattern. CONCLUSIONS: SURVIVIN is highly expressed in AL and is related to prognosis. SURVIVIN + / EPR-1 + expression pattern is more common. SURVIVIN-2B and SURVIVIN- △ EX3 may be involved in SURVIVIN-mediated apoptosis regulation.
