建立人结肠癌细胞株SW480裸鼠移植瘤模型,观察8肽生长抑素(SMS201995,SMS)对移植瘤的作用及机理。结果∶SMS组及SMS+PG(5肽胃泌素)组移植瘤的体积、重量、瘤细胞内cAMP、DNA、蛋白质含量、S期和G2M期细胞数及增殖指数均显著低于PG组及对照组,PG组显著高于对照组;而G0/G1期细胞数则显著高于PG组及对照组,PG组显著低于对照组。本实验结果提示:生长抑素具有直接抑制人结肠癌细胞株SW480移植瘤生长的作用,又可抑制胃泌素对移植瘤的促增殖作用。这种作用机理是通过抑制瘤细胞内cAMP、DNA和蛋白质的合成,从而抑制瘤细胞从G0/G1期进入到S期和G2M期。这为大肠癌患者应用生长抑素类似物进行内分泌治疗提供了实验依据
A human colon carcinoma cell line SW480 xenograft tumor model was established to observe the effect and mechanism of 8-peptide somatostatin (SMS201-995, SMS) on transplanted tumors. Results: The volume and weight of xenografts in the SMS group and SMS+PG group were significantly lower than those in the PG group and the control group. The cAMP, DNA, protein content in the tumor cells, the number of cells in the S and G2M phases, and the proliferation index were significantly lower. The PG group was significantly higher than the control group, while the number of G0/G1 phase cells was significantly higher than that of the PG group and the control group, and the PG group was significantly lower than the control group. The results of this experiment suggest that somatostatin has a direct inhibitory effect on the growth of human colon cancer cell line SW480, and it also inhibits the pro-proliferation effect of gastrin on transplanted tumors. The mechanism of action is to inhibit the synthesis of cAMP, DNA, and protein in tumor cells, thereby inhibiting the entry of tumor cells from the G0/G1 phase to the S phase and the G2M phase. This provides an experimental basis for the use of somatostatin analogues for endocrine therapy in patients with colorectal cancer
