目的研究用表皮生长因子受体(EGFR)反义寡脱氧核苷酸(ASODN)抑制人大肠癌细胞株HR8348的生长增殖,探讨大肠癌治疗的新途径.方法人工合成的EGFR反义ODN及无关对照ODN经脂质体包裹后作用HR8348细胞,采用MTT法,[3H]TdR掺入试验,细胞周期分析,裸鼠体内接种等方法测定细胞体内外增殖的变化.结果经EGFR反义ODN处理的HR8348细胞与对照组HR8348细胞相比,体外增殖速度减慢(细胞增殖抑制率达71%)、DNA合成受抑(细胞增殖指数为0361vs0784,S期细胞数为348%vs599%)、裸鼠体内成瘤率下降(25%vs100%).结论EGFR反义ODN可有效抑制大肠癌细胞的体内外生长增殖,可用于实验性肿瘤基因治疗研究
Objective To study the inhibition of growth and proliferation of human colorectal cancer cell line HR8348 by epidermal growth factor receptor (EGFR) antisense oligodeoxynucleotide (ASODN) and to explore a new approach for the treatment of colorectal cancer. Methods Synthesized EGFR antisense ODN and irrelevant control ODN were applied to HR8348 cells after liposome encapsulation. MTT assay, [3H] TdR incorporation assay, cell cycle analysis, and nude mice inoculation were used to determine the proliferation of cells in vitro and in vivo. The change. Results Compared with HR8348 cells in control group, HR8348 cells treated with EGFR antisense ODN slowed down in vitro (inhibition of cell proliferation was up to 71%) and DNA synthesis was inhibited (cell proliferation index was 0361 vs 0784, S phase). The number of cells was 348% vs599%) and the tumor formation rate in nude mice decreased (25% vs 100%). Conclusion EGFR antisense ODN can effectively inhibit the growth and proliferation of colorectal cancer cells in vivo and in vitro. It can be used for experimental tumor gene therapy research.
