目的:采用微波水解--全自动氨基酸分析法测定人工虎骨粉中总氨基酸含量。方法:样品经微波水解,采用全自动氨基酸分析法柱后衍生,测定包含羟脯氨酸、天冬氨酸、甘氨酸等总氨基酸的含量。结果:各氨基酸的线性方程R值均大于0.998,平均回收率(n=9)为98.2%。人工虎骨中甘氨酸含量最高,甘氨酸与羟脯氨酸含量的比值约为2:1。结论:该方法操作简便、省时高效,可用于人工虎骨粉中总氨基酸的含量测定。
Objective: To determine the total amino acid content of artificial tiger bone meal by microwave hydrolysis - automatic amino acid analysis. Methods: The samples were hydrolyzed by microwave and the total amino acids including hydroxyproline, aspartic acid and glycine were determined by the method of post-column derivatization with automatic amino acid analysis. Results: The linear equation of each amino acid R value was greater than 0.998, the average recovery (n = 9) was 98.2%. The highest content of glycine in artificial tiger bone, glycine and hydroxyproline content ratio of about 2: 1. Conclusion: The method is simple, time-saving and efficient, and can be used for the determination of total amino acids in artificial tiger bone meal.
