目的:探讨不同压强下持续性CO2气腹对结肠癌细胞表面黏附分子表达的影响。方法:建立体外气腹模型,选用人结肠癌细胞株SW1116,分别在不同压强CO2(6mmHg、9mmHg、12mmHg和15mmHg)以及常规条件下暴露1h。应用流式细胞技术(FACScan)检测黏附分子E鄄钙黏着素(E鄄cadherin)、ICAM鄄1、CD44、E鄄选择素(E鄄selectin)于处理后0h、12h、24h、48h及72h在结肠癌细胞表面的表达。结果:SW1116经6mmHg压强CO2气腹处理后,表面ICAM鄄1、CD44表达显著增强,9mmHgCO2气体处理处理后,E鄄选择素、E鄄钙黏着素和CD44表达显著增高,15mmHg压强CO2处理后,E鄄钙黏着素表达增强,与处理前表达水平的差异均有显著性(P<0.05),上述黏附分子的表达在各压强CO2处理后72h之前均降至处理前水平(P>0.05)。或低于处理前水平(P<0.05)。随CO2气腹压强的增高,E鄄钙黏着素、CD44和ICAM鄄1表达量逐渐降低,在处理后0h,差异均达到显著性(P<0.05)。结论:CO2气腹能对肿瘤细胞表面的黏附分子表达产生一过性双向影响。随着CO2气腹压强的增高,可抑制黏附分子表达的增强。
Objective: To investigate the effect of persistent CO2 pneumoperitoneum on the expression of adhesion molecules on colon cancer cells under different pressures. Methods: A pneumoperitoneum model was established in vitro. Human colon cancer cell line SW1116 was selected and exposed to different pressures of CO2 (6mmHg, 9mmHg, 12mmHg and 15mmHg) and routinely exposed for 1 hour. The expression of E-cadherin, ICAM-1, CD44 and E-selectin were detected by flow cytometry (FCM) at 0h, 12h, 24h, 48h and 72h Colon cancer cell surface expression. Results: The expression of ICAM-1 and CD44 on the surface of SW1116 was significantly enhanced by pneumoperitoneum CO2 of 6mmHg, and the expression of E-selectin, E-cadherin and CD44 were significantly increased after 9mmHgCO2 gas treatment. The expression of E-cadherin was significantly increased (P <0.05), and the expression of E-cadherin was lower than that before treatment (P <0.05). Or below the pre-treatment level (P <0.05). With the increase of CO2 pneumoperitoneum pressure, the expressions of E-cadherin, CD44 and ICAM-1 gradually decreased, and the difference was significant at 0h after treatment (P <0.05). Conclusion: CO2 pneumoperitoneum has a transient two-way effect on the expression of adhesion molecules on the surface of tumor cells. As CO2 pneumoperitoneum pressure increases, it can inhibit the increase of adhesion molecule expression.
