[目的]观察罗浮粗叶木醇提物体内抗鸭乙型肝炎病毒(duck hepatitis Bvirus,D-HBV)的作用。[方法]采用RT-PCR技术筛选DHBV阳性3日龄广西麻鸭60只。将麻鸭随机分为空白对照组,模型组,罗浮粗叶木醇提物高、中、低剂量组和阿昔洛韦组。用药前、用药d7、d14、d21及停药d5,取各组麻鸭静脉血,采用实时定量荧光PCR技术检测血清DHBV-DNA含量,并检测血清中AST、ALT含量和DHBsAgOD值;停药5d后取肝脏组织进行病理组织学观察。[结果]观察期内,罗浮粗叶木醇提物高、中、低剂量组与模型组病毒载量随时间的变化趋势都比较接近,差异无统计学意义(P﹥0.05);罗浮粗叶木醇提物高、中、低剂量组与模型组比较,DHBsAg差异无统计学意义(P﹥0.05);罗浮粗叶木醇提物高、中、低剂量组ALT、AST值与模型组比较,差异无统计学意义(P﹥0.05);肝脏病理检查显示各组动物肝脏病变严重程度的差异无统计学意义(P﹥0.05)。[结论]罗浮粗叶木醇提物高、中、低3个剂量组均未见明显抑制DHBV复制的作用和对DHBV引起的肝损伤具有保护作用。
[Objective] To observe the anti-duck hepatitis B virus (D-HBV) effect in vivo of Radix Octopus. [Method] RT-PCR was used to screen 60 DHBV positive 3-day-old Guangxi ducks. The ducks were randomly divided into blank control group, model group, high, medium and low dose group and acyclovir group. Before treatment, d7, d14, d21 and withdrawal d5 were taken from each group, and the serum DHBV-DNA content was detected by real-time quantitative fluorescent PCR. The contents of AST, ALT and DHBsAgOD in serum were detected. Liver tissue was taken for histopathological observation. [Result] During the observation period, the changes of viral load of high, medium and low dose group and model group were close to each other with no significant difference (P> 0.05) The levels of ALT and AST in high, medium and low dose groups were not significantly different from those in model group (P> 0.05), while those in high, medium and low dose groups were higher than those in model group Statistical significance (P> 0.05); liver pathological examination showed that the severity of liver lesions in each group of animals showed no significant difference (P> 0.05). [Conclusion] No significant inhibition of DHBV replication and no protective effect against DHBV-induced liver injury were found in the high, middle and low dose groups of the crude extract of R. roxburghii.
