目的利用基质辅助激光解析飞行(MALDI-TOF)质谱微生物检测系统对不同血清型的致泻性大肠杆菌进行分析,寻找血清型质谱分析的生物标识峰。方法将血清型可靠的5株致泻性大肠杆菌培养24 h后,进行MALDI-TOF质谱分析和特征峰聚类分析,通过对比各血清型胎指纹图谱的荷质比及相对丰度,分析血清型之间特有峰及缺失峰的差异。结果 5株致泻性大肠杆菌拥有不同的特有峰且个别相对丰度极高。EHEC亲缘关系较其他株远,峰形特异性较大。而ETEC和EAEC、EPEC和EIEC之间峰形相似度较高,亲缘关系较近。结论致泻性大肠杆菌的不同血清型的胎指纹图谱具有显著差异的生物标识峰。这些生物标识可以作为致泻性大肠杆菌血清型的质谱图谱补充,有利于完善血清学诊断相关的质谱数据库。
Objective To analyze the different serotypes of diarrheogenic E. coli with MALDI-TOF mass spectrometry and search for biomarker peaks of serotype-based mass spectrometry. Methods Five serotypes of diarrheogenic Escherichia coli were cultured for 24 h, then analyzed by MALDI-TOF mass spectrometry and characteristic peak cluster analysis. By comparing the mass-to-mass ratio and relative abundance of each serotype fetal fingerprint, Differences between unique peaks and missing peaks. Results Five diarrhea-causing Escherichia coli had different characteristic peaks and their relative abundances were extremely high. EHEC genetic relationship than other strains far, peak shape specificity. However, the peak shape similarity between ETEC and EAEC, EPEC and EIEC is relatively high and the genetic relationship is relatively close. Conclusions The fetal fingerprints of different serotypes of diarrheogenic E. coli have significantly different biomarker peaks. These biomarkers complement the mass spectra of the diarrheogenic Escherichia coli serotypes and contribute to improving the mass spectrometry database associated with serological diagnosis.
