目的:研究神经肽 Y 对血管平滑肌细胞增殖的影响。方法:应用快速自动微量 M T T比色法和荧光免疫组化定量技术观察缩血管物质神经肽 Y 对体外培养血管平滑肌细胞的 M T T比色吸光度和细胞核增殖抗原( P C N A) 表达的平均荧光值。结果:神经肽 Y 作用下血管平滑肌细胞的增殖活度和 P C N A 表达比对照组明显升高。 M T T值在 N P Y 组和对照组中分别为02626 ±00 025 和02 239 ±00 010 ,两者差异明显( P < 001) ;而 P C N A 表达的平均荧光值, N P Y 组为1 64856 ±23271 ,对照组为154271 ±20004 ,两者有差异( P < 005) 。结论:神经肽 Y 可影响血管平滑肌细胞的增殖活动;血中神经肽 Y 浓度的变化可能是高血压时血管壁结构变化的重要因素之一。
Objective: To study the effect of neuropeptide Y on the proliferation of vascular smooth muscle cells. Methods: Expressions of neurotrophin Y, a vasoconstrictor substance, on M T T colorimetric absorbance and P C N A expression in cultured vascular smooth muscle cells (VSMCs) in vitro were observed by rapid and automatic microtitration MTT assay and fluorescence immunohistochemistry The average fluorescence value. CONCLUSION: Neuropeptide Y can affect the proliferation of vascular smooth muscle cells. The change of neuropeptide Y concentration in blood may be one of the important factors in the changes of vascular wall structure in patients with hypertension.
