本文报道荧光染料双苯并咪唑Hoechst 3325 8(bi sbenzimide),应用于香菇(Lentinusedodes)、平菇(pleurotus ostreatus)、金针菇(Flammulina velutipes)、木耳(Aurtculariaauricula),毛木耳(A.polytricha)等5种食用菌菌丝体。担孢子、分生孢子核的荧光染色研究。配制染液的McIlvaine缓冲液pH值、缓冲液含0.6M MgSO_4·7H_2O、菌丝生长期、孢子贮存期亦与染色效果密切相关。我们观察到,使用插片法,爬片2/5的菌丝体,pH7.0—7.4Hoechst 3325 8染液,染色3分钟为菌丝体核荧光染色的最佳效果。新鲜的担孢子,分生孢子、使用pH 6.8含0.6 M MgSO_4·7 H:O Hoechst 3325 8染色液染色3分钟比使用pH 6.8Hoechst 3325 8的染液有明显增效作用。
In this paper, the fluorescent dye bisbenzimidazole Hoechst 3325 8 (bi-benzbenzimide) has been reported to be applied to Lentinusedodes, Pleurotus ostreatus, Flammulina velutipes, Aurtcularia auricula, A. polytricha, Edible mushroom mycelium. Study on Fluorescent Staining of Conidia and Conidium Nuclei. The pH value of the prepared McIlvaine buffer solution, buffer containing 0.6M MgSO 4 · 7H 2 O, mycelial growth period, spore storage period is also closely related with the dyeing effect. We observed that using plug method, climbing mycelium 2/5, pH7.0-7.4Hoechst 33258 dye, staining 3 minutes for mycelium nuclear fluorescence staining the best results. Fresh basidiospores and conidia, with a pH 6.8 containing 0.6 M MgSO 4 · 7 H: O Hoechst 3325 8 stain for 3 minutes than the pH 6.8 Hoechst 3325 8 stain had a significant synergistic effect.
