目的分析DNA斑点杂交技术和限制性内切酶图谱,在鉴定细菌质粒同源性中的作用。方法应用质粒图谱分析,限制性内切酶图谱分析及DNA斑点杂交等技术鉴定细菌质粒同源性。结果以质粒DNA图谱和质粒DNA限制性内切酶图谱分析只能鉴定细菌质粒数目相同的质粒同源性,而DNA斑点杂交技术还能鉴定细菌质粒数目不同的质粒同源性。结论在鉴定流行质粒的同源性方面,DNA斑点杂交技术优于上述其它方法。
Objective To analyze the DNA dot blot hybridization and restriction enzyme map in the identification of bacterial plasmid homology. Methods The plasmids were identified by plasmid map analysis, restriction endonuclease mapping and DNA dot blot hybridization. Results The plasmid DNA map and restriction map of plasmid DNA could only identify plasmid homology with the same number of bacterial plasmids. DNA dot blot hybridization could also identify plasmid homologies with different numbers of bacterial plasmids. Conclusion DNA dot blot hybridization is superior to the other methods described above in the identification of the homology of the popular plasmid.
