目的:探讨红景天苷对慢性间断性缺氧心肌细胞的影响及作用机制。方法:建立大鼠慢性间断性缺氧模型,红景天苷进行干预,实验分为假手术组、NaCl组及红景天苷组。检测心脏功能,TUNEL法检测心肌组织的心肌细胞凋亡情况。Western Blot检测心肌组织Bcl-2、Bax、caspase-3的表达情况。同时测定心肌组织caspase-3的活性。结果:与对照组比较,红景天苷干预组8周,心功能明显改善,心肌细胞的凋亡明显减少(P<0.05)。同时红景天苷干预组Bcl-2和Bax的表达比例更趋于正常化,caspase-3的表达及活性则明显减少(P<0.05)。结论:在慢性间断性缺氧中,红景天苷通过平衡Bcl-2和Bax的表达,抑制caspase-3的活性而对抗心肌细胞的凋亡,保护心功能。
Objective: To investigate the effect of salidroside on chronic intermittent hypoxic cardiomyocytes and its mechanism. Methods: Chronic intermittent hypoxia model was established in rats. Salidroside was used for intervention. The experiment was divided into sham-operation group, NaCl group and salidroside group. Cardiac function was detected by TUNEL method. Cardiomyocytes apoptosis was detected by TUNEL assay. Western Blot detection of myocardial tissue Bcl-2, Bax, caspase-3 expression. At the same time, the activity of caspase-3 in myocardium was determined. Results: Compared with the control group, salidroside intervention group for 8 weeks, cardiac function improved significantly, myocardial apoptosis was significantly reduced (P <0.05). At the same time, the expression of Bcl-2 and Bax in salidroside intervention group tended to be normalized, while the expression and activity of caspase-3 decreased significantly (P <0.05). Conclusion: In chronic intermittent hypoxia, salidroside protects cardiac function by balancing the expression of Bcl-2 and Bax, inhibiting the activity of caspase-3 against cardiomyocyte apoptosis.
