目的 :动态观察抗纤颗粒对大鼠肝纤维化模型细胞因子与肝组织胶原蛋白的影响 ,进而探讨抗纤颗粒防治肝纤维化的作用机制。方法 :将SD大鼠随机分为 10组 ,即正常对照 4W (W :Week周 )组、8W组、模型对照4W组、8W组、纤低预防 4W组、8W组、纤高预防 4W组、8W组、纤低治疗 8W组、纤高治疗 8W组。以二甲基亚硝胺(Dimethylnitrosamine ,DMN)腹腔注射建立肝纤维化模型 ,除正常对照组与模型对照组外 ,其余各组给予抗纤颗粒灌胃处理。各 4W组于实验第 30天、8W组于实验第 6 0天处死动物 ,取血作肝功能 ;TNF -α、IL - 1α、γ -IFN ;取肝组织作胶原蛋白测定。结果 :①抗纤颗粒能显著提高各期模型大鼠血清中γ -IFN含量和显著降低各期模型大鼠血清中TNF -α、IL - 1α活性及肝胶原蛋白含量 ;②TNF -α、IL - 1α与肝胶原蛋白呈显著正相关关系 (P <0 0 1,r>0 5 ) ,γ-IFN与肝胶原蛋白呈显著负相关关系 (P <0 0 1,r<- 0 5 )。结论 :抗纤颗粒预防和治疗肝纤维化的机理 ,可能与其能调节细胞因子网络有关
Objective : To dynamically observe the effect of anti-fibrosis granules on cytokines and hepatic collagen in rat liver fibrosis model, and to explore the mechanism of anti-fibrosis granules in preventing and treating hepatic fibrosis. METHODS: SD rats were randomly divided into 10 groups: normal control 4W (W Week Week) group, 8W group, model control 4W group, 8W group, fibrosis prevention 4W group, 8W group, fiber height prevention 4W group, 8W group, fiber low treatment 8W group, fiber height treatment 8W group. The hepatic fibrosis model was established by intraperitoneal injection of dimethylnitrosamine (DMN). In addition to the normal control group and the model control group, the remaining groups were treated with anti-fiber particles. The 4W group was sacrificed on the 30th day of the experiment, and the 8W group was sacrificed on the 6th day of the experiment. The blood was collected for liver function; TNF-α, IL-1α, γ-IFN; liver tissue was taken for collagen determination. RESULTS: 1 Anti-fibrin granules can significantly increase the serum γ-IFN levels and significantly reduce the activity of TNF-α, IL-1α and liver collagen in rat serum at each stage; 2TNF-α, IL - There was a significant positive correlation between 1α and liver collagen (P <0 01, r> 05), and there was a significant negative correlation between γ-IFN and liver collagen (P <0 01, r <- 05). Conclusion : The mechanism of anti-fibrosis granules in preventing and treating liver fibrosis may be related to its ability to regulate cytokine network
