目的 探索雷公藤甲素毒副作用。方法 用 0 .5mg/L ,2 .5mg/L ,10mg/L 3种浓度雷公藤甲素滴眼液滴Wistar大鼠单侧眼 ,另侧对照 ,每日 3次 ,3 0天后取角膜、眼睑病理切片 ,光镜 (LM )及电镜 (TEM)观察 ;以上述滴眼液滴单侧兔眼 ,另侧对照 ,观察评分不同时间眼部刺激 ;0 .1mg/kg雷公藤甲素注射Wistar大鼠 ,每日 1次 ,3 0天后取肝、肾病理切片观察组织变化。结果 0 .5mg/L组角膜无损害 ,2 .5mg/L组光镜无变化 ,电镜见角膜上皮部分缺损 ,10mg/L组光镜见角膜结膜上皮部分脱落 ,电镜见角膜上皮细胞核部分空泡样变 ,各组角膜基质层、后弹力层、内皮层均无损害 ;刺激反应评分为 1~ 16分 ;肝、肾细胞光镜下无明显损害。结论 0 .5mg/L雷公藤甲素滴眼液安全 ,0 .1mg/kg为雷公藤甲素安全注射量
Objective To explore the side effects of triptolide. Methods Wistar rats were randomly divided into 3 groups: unilateral eye (Wistar rats) of 0.5 mg / L, 2.5 mg / L, 10 mg / L and 10 mg / Eyelid pathological sections, light microscopy (LM) and electron microscopy (TEM) observation; the above eye drops on one side of rabbit eyes, the other side of the control, observation score eye irritation at different times; 0.1mg / kg triptolide injection of Wistar Rats, once a day, 30 days after liver and kidney pathological changes were observed. Results There was no damage to the cornea in 0.5mg / L group, no changes in light microscope in 2.5mg / L group, partial corneal epithelial defect in electron microscope, partial detachment of corneal conjunctival epithelium in 10mg / L group, and partial vacuolation of corneal epithelium in electron microscope The changes of the corneal stroma, the posterior elastic layer and the endothelium were not damaged in all groups. The irritation response score was 1 ~ 16 points. There was no obvious damage in the liver and kidney cells under light microscope. Conclusion 0 .5mg / L triptolide eye drops safety, 0.1mg / kg triptolide safe injection volume
