目的:观察鱼腥草合剂醇提液和水提液对糖尿病大鼠肾脏结缔组织生长因子和转化生长因子表达的影响,探讨其防治糖尿病肾病的发病机制以及与炎症因子的相关性。方法:采用左侧肾切除手术+高糖高脂饲料+小剂量链脲佐菌素(STZ,30 mg.kg-1)腹腔注射三联法造成2型糖尿病肾病大鼠模型。雄性Wistar大鼠被随机分为正常组、模型组、西药对照组(氯沙坦组和罗格列酮组)、鱼腥草合剂醇提组(高剂量组和低剂量组)、鱼腥草合剂水提组(高剂量组和低剂量组)。采用放免法测定尿白蛋白(UAlb);采用免疫蛋白印迹(Western-blot)法观察肾组织结缔组织生长因子(CTGF)和转化生长因子-β1(TGF-β1)的表达量。结果:治疗8周后,各给药组糖尿病大鼠多饮、多食和体重降低状况均明显改善。合剂醇提组在改善糖尿病大鼠肾损伤、减少24 h尿蛋白、UAlb,抑制肾组织CTGF和TGF-β1表达量方面均优于西药对照组。结论:鱼腥草合剂可减少糖尿病大鼠尿蛋白、UAlb和内生肌酐清除率,降低肾脏CTGF和TGF-β1的表达。
Objective: To observe the influence of Houttuynia mixture alcohol extract and water extract on the expression of connective tissue growth factor and transforming growth factor in diabetic nephropathy rats, and to explore its pathogenesis of diabetic nephropathy and the correlation with inflammatory factors. Methods: A rat model of type 2 diabetic nephropathy was established by intraperitoneal injection of the left nephrectomy plus high-sucrose and high-fat diet + low dose streptozotocin (STZ, 30 mg.kg-1). Male Wistar rats were randomly divided into normal group, model group, western medicine control group (Losartan group and rosiglitazone group), Houttuynia mixed alcohol group (high dose group and low dose group), Houttuynia Mixture water extraction group (high dose group and low dose group). Urinary albumin (UAlb) was determined by radioimmunoassay. Expression of connective tissue growth factor (CTGF) and transforming growth factor-β1 (TGF-β1) in renal tissue was detected by Western-blot. Results: After 8 weeks of treatment, the diabetic rats in each treatment group were significantly more likely to drink more, eat more and lose weight. The combination of alcohol extract group is better than western medicine control group in improving renal injury, reducing 24 h urinary protein, UAlb, inhibiting renal CTGF and TGF-β1 expression in diabetic rats. Conclusion: Houttuynia mixture can reduce urinary protein, UAb and creatinine clearance in diabetic rats and decrease the expression of CTGF and TGF-β1 in kidney.
