【目的】观察黄连解毒汤对动脉粥样硬化(AS)大鼠主动脉组织中单核细胞趋化蛋白-1(MCP-1)及其特异性受体CCR2 mRNA表达的影响。【方法】选用SD大鼠,随机分为正常对照组,模型组,黄连解毒汤低、中、高剂量组(中药组,剂量分别为2.7、5.3、10.6 g.kg-1.d-1),阿托伐他汀组(剂量为1.8 mg.kg-1.d-1)。除正常对照组外,其他组均采用高脂饮食法复制AS模型,造模第6周开始给药,连续4周。取各组大鼠主动脉采用实时荧光定量PCR法检测各组MCP-1/CCR2 mRNA表达。【结果】与正常对照组比较,模型组主动脉MCP-1/CCR2 mRNA表达显著升高(P<0.05或P<0.01);与模型组比较,黄连解毒汤各剂量组MCP-1/CCR2 mRNA表达均显著降低(P<0.05或P<0.01),且呈一定的剂量效应关系,MCP-1与CCR2两者表达呈正相关。【结论】黄连解毒汤治疗AS的作用可能与其能下调MCP-1、CCR2 mRNA在主动脉的表达有关。
【Objective】 To observe the effect of Huanglian Jiedu Decoction on the expression of monocyte chemoattractant protein-1 (MCP-1) and its specific receptor CCR2 mRNA in the aorta of rats with atherosclerosis (AS). 【Methods】 SD rats were randomly divided into normal control group, model group, Huanglian Jiedu Decoction low, middle and high dose groups (Chinese medicine group, the dosage was 2.7, 5.3, 10.6 g.kg-1.d-1 respectively). In the atorvastatin group (dose 1.8 mg.kg-1.d-1). In addition to the normal control group, the other groups were all duplicated by the high-fat diet AS model, the model began to be administered 6 weeks, for 4 weeks. The expression of MCP-1/CCR2 mRNA in each group was detected by real-time fluorescence quantitative PCR. 【Results】 Compared with the normal control group, the expression of MCP-1/CCR2 mRNA in the aorta of the model group was significantly increased (P<0.05 or P<0.01). Compared with the model group, the MCP-1/CCR2 mRNA was observed in each group of Huanglian Jiedu Decoction. The expression was significantly decreased (P<0.05 or P<0.01), and showed a certain dose-effect relationship. There was a positive correlation between the expression of MCP-1 and CCR2. 【Conclusion】 The effect of Huanglian Jiedu Tang in the treatment of AS may be related to its ability to down-regulate the expression of MCP-1 and CCR2 mRNA in the aorta.
