目的探讨丹蛭降糖胶囊对糖尿病大鼠血清基质细胞衍生因子-1α(SDF-1α)及细胞周期调节蛋白P27的影响。方法 SD大鼠48只,分为正常对照组,模型组,丹蛭降糖胶囊(DJC)高、低剂量组,吡格列酮组。除正常对照组外,其余动物给予高脂饲料喂养4周,按35μg/g给大鼠一次性腹腔注射链脲佐菌素(STZ)溶液造模。造模成功后,连续灌药8周,抽取腹主动脉血,采用双抗体一步夹心法测定血清SDF-1α和P27水平,并测定血糖及糖化血红蛋白(HbA1c)。结果与正常组比较,模型组SDF-1α水平下降明显(P<0.01),而P27表达升高(P<0.01);与模型组比较,丹蛭降糖胶囊高、低剂量组与吡格列酮组SDF-1α水平显著升高(P<0.01),P27水平明显下降(P<0.01);3个给药组之间比较,SDF-1α、P27差异均无统计学意义(P>0.05)。与正常组比较,模型组血糖、HbA1c显著升高,用药后丹蛭高、低剂量组血糖、HbA1c均发生明显下降。相关性分析显示,血糖与SDF-1α呈显著负相关(P<0.01),与P27呈显著正相关(P<0.01)。结论丹蛭降糖胶囊能够使糖尿病大鼠血清SDF-1α水平升高,抑制糖尿病大鼠P27的高表达,延缓糖尿病肾脏病变,保护肾脏功能,有明确的降低血糖及HbA1c的作用。
Objective To investigate the effect of danzhijiangtang capsule on serum SDF-1α (SDF-1α) and cell cycle regulatory protein P27 in diabetic rats. Methods Forty-eight SD rats were divided into normal control group, model group, Danzhijiangtang capsule (DJC) high and low dose group and pioglitazone group. Except for the normal control group, the other animals were given high-fat diet for 4 weeks, and the rats were given a single intraperitoneal injection of streptozotocin (STZ) solution at 35 μg / g. After successful model making, continuous aortic perfusion was performed for 8 weeks. Abdominal aorta blood was drawn. Serum SDF-1α and P27 levels were measured by double-antibody one-step sandwich method. Blood glucose and HbA1c were measured. Results Compared with the normal control group, the level of SDF-1α in the model group decreased significantly (P <0.01) and the expression of P27 increased (P <0.01). Compared with the model group, (P <0.01), while the level of P27 was significantly decreased (P <0.01). There were no significant differences in SDF-1α and P27 between the three treatment groups (P> 0.05). Compared with the normal group, the blood glucose and HbA1c of the model group were significantly increased, and the blood glucose and HbA1c of the high and low dose lentils decreased significantly after treatment. Correlation analysis showed that there was a significant negative correlation between blood glucose and SDF-1α (P <0.01), and positive correlation with P27 (P <0.01). Conclusion Danzhi Jiangtang Capsule can increase serum SDF-1α in diabetic rats, inhibit the expression of P27, delay the diabetic nephropathy, protect renal function, and obviously decrease the blood glucose and HbA1c in diabetic rats.
