目的研究新生大鼠缺氧缺血性脑损伤后FADD的表达水平变化及头孢曲松对其表达的影响。方法将7日龄新生大鼠随机分为假手术组,生理盐水对照组和头孢曲松组,按照观察时间点不同进一步分为6h、12h、24h、3d、5d、7d共6个亚组,每个亚组8只,头孢曲松组于造模后连续3d每天腹腔注射头孢曲松(200mg/kg),每天1次,假手术组及生理盐水组按同样方法注射等量生理盐水。Western blot法和免疫组化法检测新生大鼠脑组织中FADD的表达。结果假手术组各个时间点FADD表达量均相对较少,生理盐水对照组和头孢曲松组FADD表达水平在各个时间点均较假手术组有明显升高,差异有统计学意义(P<0.05);头孢曲松组FADD在各个时间点表达水平均低于对照组,差异有统计学意义(P<0.05)。结论头孢曲松可能通过降低缺氧缺血性脑损伤新生大鼠脑组织FADD表达,从而发挥神经保护作用。
Objective To investigate the changes of FADD expression in neonatal rats after hypoxic-ischemic brain damage and the effect of ceftriaxone on it. Methods 7-day-old neonatal rats were randomly divided into sham operation group, saline control group and ceftriaxone group. According to the different time points, 6 newborn rats were further divided into 6 subgroups at 6h, 12h, 24h, 3d, 5d and 7d, In each subgroup, ceftriaxone group was given intraperitoneal injection of ceftriaxone (200mg / kg) once a day for 3 consecutive days after modeling. The rats in sham operation group and normal saline group were given the same amount of normal saline by the same method. The expression of FADD in brain tissue of neonatal rats was detected by Western blot and immunohistochemistry. Results The expression of FADD in sham operation group was relatively lower at all time points. The expression of FADD in saline control group and ceftriaxone group was significantly higher than that in sham operation group at each time point (P <0.05 ). The levels of FADD in the ceftriaxone group were lower than those in the control group at each time point, the difference was statistically significant (P <0.05). Conclusion Ceftriaxone may play a neuroprotective role by decreasing FADD expression in neonatal rats with hypoxic-ischemic brain damage.
