以美国大灵芝为出发菌株,利用N+离子注入技术选育出一株遗传性状稳定的灵芝多糖高产菌株。结果表明:在20 KeV2、×1016ions/cm2的注入条件下灵芝菌株G13的胞外多糖的产量为2.71 g/L,比对照菌株提高了15.32%。采用单因素法和正交实验法得到的培养基组分质量浓度优化组合为:葡萄糖45 g/L,玉米粉40 g/L,黄豆粉10 g/L,酵母膏0.5 g/L,VB125μg/L,KH2PO40.5 g/L,MgSO4.7H2O 0.1 g/L。进行分批发酵实验,发酵120 h灵芝胞外多糖的产量达到3.38 g/L。
Taking the American ganoderma lucidum as the starting strain, a high yielding strain of ganoderma lucidum polysaccharide with stable genetic character was bred by using N + ion implantation technology. The results showed that the exopolysaccharide yield of Ganoderma lucidum strain G13 was 2.71 g / L under the injection condition of 20 KeV2 × 1016ions / cm2, which was 15.32% higher than that of the control strain. The optimal combinations of the medium components were as follows: glucose 45 g / L, corn flour 40 g / L, soybean flour 10 g / L, yeast extract 0.5 g / L, VB125 μg / L, KH2PO40.5 g / L, MgSO4.7H2O 0.1 g / L. Batch fermentation experiments, 120 h fermentation of Ganoderma lucidum polysaccharide production reached 3.38 g / L.
