为了检测 Rb,p53,p16基因产物在非小细胞肺癌(NSCLC)中的表达特征,采用免疫组化和图像定量相关分析技术比较了 Rb,p53与 p16的表达情况及相互关系。结果11例 Rb 表达阴性的肿瘤,p16蛋白表达皆为强阳性,而30例 Rb 表达阳性程度较高(++~+++)的标本中,23例显示 p16表达阴性或低水平表达,图像定量相关分析显示 Rb 失活与 p16表达之间存在负相关关系。提示 NSCLC 发生发展的分子遗传机制可能涉及 p16及 Rb 基因在内的细胞周期 G_1期负反馈调节通路异常。
In order to detect the expression characteristics of Rb, p53 and p16 gene products in non-small cell lung cancer (NSCLC), immunohistochemistry and image quantitative correlation analysis techniques were used to compare the expression and relationship of Rb, p53 and p16. Results The expression of p16 protein was strongly positive in 11 cases with negative Rb expression, and in 30 cases with high Rb positive expression (++ - +++), 23 cases showed negative or low expression of p16. Quantitative correlation analysis revealed a negative correlation between Rb inactivation and p16 expression. It is suggested that the molecular genetic mechanism of the development of NSCLC may be related to abnormalities in the G1 phase negative feedback regulatory pathway of the p16 and Rb genes.
