目的探讨microRNA-424-5p(miR-424-5p)在辐射诱导的A549细胞侵袭和迁移中的作用。方法采用茎环RT-PCR检测辐照后A549细胞中miRNA的表达;Transwell小室实验检测A549细胞侵袭和迁移能力。结果 4GyX射线照射诱导了A549细胞中miR-424-5p的表达;过表达miR-424-5p后,与对照组相比,A549细胞侵袭和迁移能力明显增强;导入外源性miR-424-5p抑制物后进行4 GyX射线照射,A549细胞侵袭和迁移能力明显减弱。结论 4 GyX射线诱导了miR-424-5p的表达并促进了A549细胞的侵袭和迁移。本研究为未来进一步研究其分子作用机制奠定基础。
Objective To investigate the role of microRNA-424-5p (miR-424-5p) in radiation-induced invasion and migration of A549 cells. Methods The stem-loop RT-PCR was used to detect the expression of miRNA in A549 cells after radiation. The invasion and migration of A549 cells were detected by Transwell chamber assay. Results 4Gy X-ray irradiation induced the expression of miR-424-5p in A549 cells. Compared with the control group, the invasion and migration of A549 cells were significantly enhanced after overexpression of miR-424-5p. The exogenous miR-424-5p Inhibition of 4 Gy X-ray irradiation, A549 cell invasion and migration was significantly weakened. Conclusion 4 Gy X-ray induced the expression of miR-424-5p and promoted the invasion and migration of A549 cells. This study lays the foundation for further research on the molecular mechanism of action in the future.
