本文应用肉芽组织囊试验,研究了醋酸棉酚对实验动物成纤维细胞的减数分裂、SCE 和细胞点突变的影响。随醋酸棉酚剂量的增加,离体成纤维细胞有丝分裂指数,从7.46%下降至0.80%、SCE 平均每一细胞从4.33次上升到20次、细胞突变率从7.20×10~(-6)上升到12.05×10~(-6).各剂量组与对照组间均具显著性差异(P<0.01)。结果表明,低剂量醋酸棉酚即可导致实验动物离体细胞DNA 初级结构的损伤和诱发活体动物细胞突变。
In this paper, the granulation tissue test was used to study the effects of gossypol acetate on the meiosis, SCE and cell-point mutation in experimental animal fibroblasts. The mitotic index of isolated fibroblasts decreased from 7.46% to 0.80% with the increase of the dose of gossypol acetate, and the average SCE increased from 4.33 to 20 times per cell. The mutation rate of cells increased from 7.20 × 10 ~ (-6) To 12.05 × 10 ~ (-6), there was a significant difference between each dose group and control group (P <0.01). The results showed that low dose of gossypol acetate can lead to DNA damage primary structure of experimental animals in vitro and induced mutations in living animal cells.
