目的探讨慢性应激状态下大鼠不同脑区pCREB表达水平的变化特征。方法体质量185~255 g雄性Wistar大鼠30只,随机分为对照组(C组15只)和实验组(T组15只)。T组大鼠单只分笼喂养,给予21 d的不可预见性综合刺激;C组大鼠分5只/笼喂养,不给予任何刺激。第22天脑组织取材、切片并进行pCREB的免疫组织化学测定;实验前后分别进行旷场分析法和体质量测定。结果实验后的T组与C组相比:体质量减轻(P<0.05);中央格停留时间延长(P<0.01);水平穿越格数减少(P<0.01);直立次数减少(P<0.01);修饰次数减少(P<0.01);粪便粒数增多(P<0.01)。2组在海马CA3区和齿状回(DG)均有阳性表达,但T组的平均光密度值均比C组显著减少(P值均<0.01)。结论慢性综合应激减少大鼠的探究行为,抑制修饰行为,增加排便量;慢性应激后,大鼠海马神经元pCREB表达显著下降。
Objective To investigate the changes of pCREB expression in different brain regions of rats under chronic stress. Methods Thirty male Wistar rats weighing 185-255 g were randomly divided into control group (15 in C group) and experimental group (15 in T group). The rats in T group were fed only in cage, giving unplanned comprehensive stimulation for 21 days. Rats in C group were fed with 5 rats / cage without any stimulation. On the 22nd day, the brain tissues were taken, sectioned and immunohistochemically detected by pCREB. Open-field analysis and body mass measurement were performed before and after the experiment. Results After the experiment, compared with group C, the body weight was reduced (P <0.05); the stay time in the central grid was prolonged (P <0.01); the number of horizontal crosses was decreased (P <0.01) ); The number of modification decreased (P <0.01); the number of fecal particles increased (P <0.01). There was a positive expression in hippocampal CA3 area and dentate gyrus (DG), but the average optical density value in group T was significantly lower than that in group C (all P <0.01). Conclusion Chronic comprehensive stress can reduce the exploring behavior of rats, inhibit the modification behavior and increase the amount of defecation. After chronic stress, the expression of pCREB in hippocampal neurons decreased significantly.
